An alternative pathway for the processing of Okazaki fragments

A novel molecular defect in ARH3-mutated neurodegenerative disease
We have found that whilst the absence of ARH3 glycohydrolase does not impede DNA repair, instead it leads to the persistence of aberrant ADP-ribose chromatin “scars” at sites of previous PARP-dependent DNA repair events. [pubmed] [doi]
New insights into PNKP-mutated neurological disease
We implicate reduced rates of DNA single-strand break repair, not DNA double-strand break repair, as the source of both neurodevelopmental and neurodegenerative pathology in PNKP-mutated disease. We also point to the extent of this reduction as the determinant of disease severity. [pubmed] [doi]
